ABSTRACT


Background: Piper betle Linn facilitate the tumor antigen initiation on immunosurveilance cells. Phylanthus niruri Linn improve the effectiveness of tumor cell of Adenocarcinoma mammae destruction. This study was aimed to prove any change of histopathological appearance of Adenocarcinoma mammae in C3H mice that give by Piper betle Linn and Phylanthus niruri Linn extracts.
Methods: This was a post-test only control group design, using 10 C3H mice that fulfilled the inclusion criteria, fed with 511 standards and SGM 1 milk ad libitum. At, the second weeks, all were inoculated by Adenocarcinoma mammae, 0,2 ml sub cutaneously, divided into two groups. At the third weeks, the control group just only give by standard dietary, but the treatment group added by Piper betle Linn and Phylanthus niruri Linn extracts (25 mg/ml) per sonde daily. At the 7th weeks, all samples were terminated, the mammae were processed histopathologically, and examine using the Eltson modified of Bloom-Richardson score (B-R). The data were test by Kolmogorov-Smirnov, as well as t-test dan Mann-Whitney.
Results: All grading score of sample from K and P groups showed B-R=9 (grade III), except one sample in P group (B-R=7 or grade II). The normality distribution test of Kolmogorov-Smirnov were abnormal (p<0,05). The Mann-Whitney test has no significant different between histologic grading in K and P groups, p=0,317 (p>0,05). The tumor weight distribution data was normal (p>0,05), and t-test has significant different between K and P groups, p=0,048 (p<0,05).
Conclusions: The C3H mice that inoculated by Adenocarcinoma mammae and give by Piper betle Linn and Phylanthus niruri Linn extracts were not have significant different in histopathological pattern of Adenocarcinoma mammae.

ABSTRAK

Latar belakang: Daun sirih dapat mempermudah pengenalan antigen sel tumor pada sel imunosurveilans. Meniran mengefektifkan penghancuran sel tumor Adenocarcinoma mammae. Penelitian ini bertujuan untuk membuktikan adanya perubahan gambaran histopatologi Adenocarcinoma mammae pada mencit C3H yang diberi ekstrak sirih dan meniran.
Metode: Desain penelitian eksperimental ini adalah post test only control group, memakai 10 ekor C3H yang memenuhi kriteria inklusi, diaklimatisasi, diberi pakan standar 511 dan susu SGM 1 secara ad libitum. Pada minggu kedua seluruh mencit diinokulasi sel tumor Adenocarcinoma mammae dengan dosis 0,2 ml sub cutan aksila kiri, dibagi menjadi 2 kelompok. Mulai minggu ke-3 sampai keenam, kelompok kontrol (K) diberi diet standar. Kelompok perlakuan (P) diet standar, ekstrak sirih dan meniran (dosis 25 mg/ml) per sonde sekali sehari. Pada minggu ke-7, seluruh mencit diterminasi, diambil jaringan tumor payudara, diproses histopatologis, dinilai berdasarkan kriteria Bloom-Richardson yang dimodifikasi Eltson (B-R). Uji normalitas data dengan Kolmogorov-Smirnov, dilanjutkan analisis t-test dan Mann-Whitney.
Hasil: Seluruh sampel dari kelompok K dan P memperlihatkan skor grading B-R=9 (grade III), kecuali satu sampel kelompok P (P-2) yang memperlihatkan nilai B-R=7 (grade II). Uji normalitas data dengan Kolmogorov-Smirnov didapatkan distribusi tidak normal (p<0,05). Uji Mann-Whitney terbukti tidak ada perbedaan signifikan antara grading histologi kelompok kontrol (K) dan perlakuan, p=0,317 (p>0,05). Distribusi data berat tumor normal (p>0,05), dan t-test mendapatkan ada perbedaan signifikan antara kelompok kontrol (K) dan perlakuan, p=0,048 (p<0,05).
Kesimpulan: Pemberian ekstrak sirih dan meniran tidak memberikan perubahan bermakna terhadap gambaran histopatologi tumor Adenocarcinoma mammae mencit C3H, namun menunjukkan adanya penurunan bermakna pada berat tumor.
