ABSTRACT

Background: Cyclea barbata L. Miers (cincau hijau) may increase tumor cells necrosis significantly, though no studies about the potential effect on skin cancer, besides the incidence rate of skin cancer is still high. This study was aimed to know the effect of C. barbata L. Miers leaves extract on the histopathology epidermal changes of C3H that exposed with carcinogen.
Methods: This was a post test only control group design study on 20 C3H mice, randomly labeled into 4 groups. Group K1, standard dietary for three weeks; K2, standard dietary plus 1.62 ml C. barbata L. Miers leave extract per sonde every 3 weeks, than were exposed with DMH for 4 months; P2, standard dietary, DMH exposure for 4 months, plus 1.62 ml C. barbata L. Miers. In the end of study, the histopathology examination was made. The data were tested using Kruskal-Wallis and Mann-Whitney.
Results: Mean score of histopathologic epidermal changes on K2 was highest (1.98±1.10); while K1 (0.06±0.24) was the lowest. P1 and P2 higher than K1, but lower than K2. P1 higher than P2. Kruskal-Wallis test showed the significantly differences on all groups. Mann-Whitney test showed the significant differences between K1 v sK2, K1 vs P1, K2 vs P2, and P1 vs P2; however between K1 vs P2 and K2 vs P1 were not found any significant differences (p>0,05).
Conclusion: There were no significantly differences on the histopathology epidermal changes between group that added by C. barbata L. Miers leave extract with the group only exposed by DMH before.

ABSTRAK

Latar belakang: C. barbata L. Miers dapat meningkatkan nekrosis sel tumor secara signifikan, namun potensi terhadap kanker kulit belum ada penelitian. Penelitian ini bertujuan mengamati perbedaan perubahan epidermis mencit C3H yang dipapar karsinogen ter dan diberi ekstrak daun C. barbata L. Miers.
Metode: Penelitian eksperimental post test only control group design dengan 20 mencit jantan C3H, dibagi random menjadi kelompok; K1, diet standar selama 3 minggu; K2, diet standar dan paparan ter pada kulit punggung setiap hari selama 4 bulan; P1, diet standar, diberi 1,62 ml ekstrak daun C. barbata L. Miers per sonde setiap hari selama 3 minggu, kemudian paparan ter setiap hari selama 4 bulan; P2, diet standar, paparan ter setiap hari selama 4 bulan, diberi 1,62 ml ekstrak daun C. barbata L. Miers. Setelah perlakuan berakhir, dilakukan pemeriksaan histopatologi dengan skoring. Analisis data dengan uji Kruskal-Wallis dan Mann-Whitney.
Hasil: Rerata skor K2 tertinggi dibanding kelompok lain (1,98±1,10); sedangkan rerata skor terendah pada K1 (0,06±0,24). Rerata skor P1 maupun P2 lebih besar daripada K1, tetapi lebih kecil daripada K2. Rerata skor P1 lebih besar daripada P2. Uji Kruskal-Wallis terdapat perbedaan sangat bermakna, Mann-Whitney ada perbedaan sangat bermakna antara K1 dengan K2, K1 dengan P1, K2 dengan P2, dan P1 dengan P2; sedangkan antara K1 dengan P2 dan K2 dengan P1 tidak ada perbedaan bermakna (p>0,05).
Kesimpulan: Tidak terdapat perbedaan yang bermakna pada perubahan epidermis mencit C3H antara kelompok yang diberi ekstrak daun C. barbata L. Miers sebelum dipapar ter dengan kelompok yang hanya dipapar ter.
